Default Quick question about testing electricals

I need to test my TFI module and other components with a multimeter.

Do I disconnect the negative battery terminal whilst doing this.

I tried it before and got no reading from the multimeter (probed pins 4 and 5 of TFI connector). Does the battery have to be on?
